All Fun and q-PCR

Welcome back fish friends,

Happy 4th of July all you fish lovers!!! Hopefully you got to relax and have a little fun, we sure did.

 

Weekly Updates:

This is week 7 of the 10 week summer research program the Weicksel Lab is involved in.

Monday and Tuesday we didn't meet as a lab because of the 4th of July holiday. We had a little extra time off to swim around and have fun!

On Wednesday, we ran our q-PCR from our 3C protocol last week. It looked like some of the pieces amplified but not all of it, so it looks like we have some troubleshooting to do. Then we prepared a gel for our q-PCR. After we ran the gel we did a gel extraction to send off for sequencing to find out what exactly was going on in our q-PCR.

Thursday, we mated some fish because we want more embryos. Sadly, the fish didn't give us a ton of embryos, so looks like more matings will happen next week. We fixed their embryos for 3C again. We also set up another q-PCR to test the primers and our 3C libraries. Turns out it was a little weird so we are back to trouble shooting.

Friday, brought yet another q-PCR (girl, you crazy... we know we know). Yes we set up another q-PCR. Fingers crossed this one actually works and we can get some results. The q-PCR turned out to be pretty good but some trouble shooting still needs to be worked out. It's all part of the process, figuring out how each step of the process needs to go so we can get accurate and unbiased results.
